The College of Dentistry was first established as a unit of the Philippine College of Commerce and Business Administration in 1948 in a building at the main campus facing S. Loyola Street. The College was granted government recognition on July 1, 1950. In July 1964, the College of Dentistry was transferred from the UE Main/Recto to Quezon City where it shared the campus with the UERMMC College of Medicine, School of Nursing, and the UE Research Center.

In June 1988, to give way to the University's plan to add new curricular offering in the UERM College of Medicine and for the College of Dentistry to be able to expand its facilities in order to keep abreast with the new trends and developments in dental medicine, a gradual transfer of the College back to UE Manila campus was initiated. It started with freshmen dental proper students enrolled in SY 1988-1989 who would hold the classed in the new Dental Building (formerly Science Building) that had undergone extensive renovations and refurbishing to meet the increasing demand for modern technology in dental education. The new site was formally inaugurated on September 16, 1991 and has provided more room for the massive expansion of the College's Laboratory and Clinical facilities where all instructional and clinical activities are conducted. In addition, as of SY 2004, the hospital internship of senior dental clinicians are being held again at the UERMMMC. The current dean of the College of Dentistry is Dr. Diampo J. Lim, a Magna Cum Laude graduate of UE, class of 1960.

Many may ask why Dental Medicine Interns are in UERMMMC. The answer cannot be simply heard but are invinsibly spoken by every oral cavity of each person...that the mouth is still an essential part of the human body. A dentoalvolar abscess can kill a person so much ones killer smile.
